一、指南说明
“十一五”863计划先进制造技术领域“生命科学微量样品自动化操作设备”重点项目是依据《国家中长期科学和技术发展规划纲要》和《国家高技术研究发展计划(863计划)“十一五”发展纲要》的任务要求设置的。
生命科学是当今科技中最为活跃的研究领域之一,克隆技术、基因与蛋白质等研究是生命科学研究领域的前沿方向,其发展和进步依赖于先进的实验方法、手段和仪器设备。目前生命科学仪器与设备向自动化、通量化方向发展, 国内需求量大,但主要设备依赖进口。因此,研究具有我国自主知识产权的自动化、通量化生命科学自动化仪器与设备,对打破国外技术垄断、促进我国生命科学发展及设备的产业化具有重要的意义。
项目总体目标:针对生命科学研究向精细化、规模化、通量化发展的趋势和我国在生命科学研究与应用中对高通量、自动化、精密化仪器与设备的迫切需求,围绕生命科学中的体细胞克隆、基因与生物试样处理、蛋白质提取与高通量筛选中的自动化操作与处理,基于机器人与自动化技术、生物医学技术、微纳制造技术等高新技术,开发出若干具有市场竞争能力的生命科学微量样品自动化操作设备,实现产品定型、小批量生产和示范应用,为我国生命科学研究达到国际先进水平提供重要的手段,并为具有自主知识产权的生命科学仪器与设备的产业化奠定基础。
项目主要任务:围绕生命科学微量样品自动化操作的实际需求,重点研究开发蛋白质自动化提取与多孔板微阵列制备、生物微量试样处理、生物组织微操作等自动化设备,解决生物组织分离、超微量显微注射、微量生物试剂分配、蛋白质自动化提取与高通量筛选、信息处理、系统集成与示范应用等关键技术。
项目申请要求:申请单位根据项目指南发布的全部研究内容和有关要求自行联合国内优势单位进行申请,而不能只针对项目部分目标和任务进行申请,鼓励产学研结合。由参与单位自行组成项目申请团队(同一个人只能参加一个申请团队),并提出项目牵头单位和项目召集人,由项目牵头单位具体负责项目申请。本项目采取评审择优的方式确定项目和课题承担单位。

课题1、蛋白质自动化提取与多孔板微阵列制备系统 1.主要研究内容
针对生命科学中蛋白质功能研究的需求,围绕集成化、通量化、精确化的蛋白质自动化操作,研究开发蛋白质冷冻、研磨、离心、清洗等蛋白质自动化提取和多孔板微阵列制备系统,攻克蛋白质提取操作的自动化与集成化、微量生物样品的分配、微流体控制、系统集成与示范应用等关键技术。
2.主要考核指标
① 蛋白质自动化提取系统

能实现研磨、分离、清洗的自动化连续操作; 可处理样品:动物组织、微生物、植物组织; 蛋白质回收率: >90%; 结果稳定性CV:<5%;
② 多孔板微阵列制备系统

微阵列密度: 10000dots/cm2。
③ 研制出实用化蛋白质自动化提取与多孔板微阵列制备系统工程样机,并实现15 台以上的小批量生产与应用。

针对生命科学中病理医学研究、克隆技术研究、基因及蛋白质工程研究对特定细胞、特定组织等目标选择性自动化显微操作的需求,研究开发生物细胞显微注射和生物组织显微切割设备,攻克三维显微视觉、微量精确注射、超声显微分离、系列化作业工具设计等关键技术。
